Science requires proper controls. BAPTA evoked MCL-1 downregulation, so "Ca2+ drives MCL-1 abundance"? It turned out that TF-BAPTA (Kd 65 µM, far off 100 nM basal [Ca2+]) too evoked MCL-1 decline! So, Ca2+ chelation was not the cause. Our journey led to direct PFKFB3 inhibition!

#calcium #BAPTA #controls #PFKFB3 #MCL1 #cell_death @EuroCalcium It wasn't easy and risky working on an "off-target". But, we found it worthwhile to properly interpret findings as BAPTA is used in >1000s studies but also learning new things such as PFKFB3 linking to MCL-1.
