The receptor is “captured” bound by GABA but with the 9’ activation pore gate closed. This is consistent with the low probability of opening that alpha-beta channels show (versus alpha-beta-gamma channels), a natural means to limit their impact on blocking nerve signalling.
The endogenous neuromodulator, zinc, is shown to block the GABA-A receptor pore at a high sensitivity site on the extrasynaptic alpha-beta receptor type.
The neurotoxin alpha-Cobratoxin which causes paralysis by acting on nicotinic acetylcholine receptors is shown here bound to a GABA-A receptor to stabilize the “resting” (closed) state.
